DESCRIPTION

The person who fills this position will assume work expressing recombinant proteins in bacteria and in cell lines such as CHO cells. Many of these recombinant proteins will be studied in the laboratory as candidate therapeutics for inflammatory diseases. Handling of DNA constructs will be important. Kit-based site-directed mutagenesis will be used regularly. Understanding how to examine newly expressed recombinant proteins by methods such as SDS-PAGE, immunoblotting, and protein-binding assays will be important. The successful candidate will be encouraged to perform independent experiments using purified proteins and cells in culture to analyze protein activity. Analysis of cell-signaling responses in cultured cells is accomplished by a variety of methods that may be learned. Methodical preparation and maintenance of reagents is essential. Strong organizational and record-keeping skills are necessary. Use of statistical computer software packages to analyze data will be important. There will be an excellent opportunity to publish studies as an author or co-author, dependent on successful contributions to laboratory products. A team-based and supportive approach to scientific research is strongly encouraged.

MINIMUM QUALIFICATIONS

  • Theoretical and practical knowledge of cellular and molecular biology, protein biochemistry, tissue culture, and bacteria, or combination of education and experience.

  • Ability to perform standard repetitive techniques that may be required in research protocols related to the laboratory. Ability to interpret and apply protocols in conducting research. Skill to troubleshoot adaptations of experimental protocols.

  • Knowledge of and experience working with plasmids, preparation of solutions/media, bacteriology & molecular cloning..

  • Maintain multiple different eukaryotic cell lines including freezing, thawing, storage, and plating cells in preparation for experiments. Experience with sterile techniques, trouble shooting of contamination problems, and care of incubators and liquid nitrogen freezers..

  • Theoretical and practical knowledge of molecular biology techniques in extracting DNA/RNA, PCR, RT-PCR.

  • Knowledge of protein chemistry/purification, gel electrophoresis and transfers, various gel and membrane stains, western blots, and densitometry and analysis on resulting data.

  • Good organizational skills, and the ability to organize and maintain a clean laboratory, maintain inventory and order supplies..

  • Ability to prioritize, work independently and efficiently.

  • Ability to maintain effective working relationships with other staff members, students, faculty and scientists in a multi-cultural lab.

  • Knowledge of laboratory safety including biohazard handling and disposal techniques. Demonstrated ability to follow established Safety and Biohazardous Material Guidelines and implement measures to ensure compliance and maintain associated records.

  • Willing to comply with UCSD policies and procedures and to stay informed and comply with basic safety rules and issues.

  • Ability to make independent decisions, analyzing situations, problem solving, and initiating action for laboratory maintenance.

  • Organizational skills to order and maintain lab reagents and supplies, and keep accurate lab records and computer files. Experience in ordering, maintaining, trouble-shooting, and calibrating general laboratory equipment (e.g. centrifuges, incubators, etc).

  • Experience with the operation of a personal computer and software applications, such as Microsoft Office (Word, Excel, PowerPoint), Photoshop, Acrobat.

  • Willingness to learn new theory, techniques, and tasks and share knowledge.

  • Proven ability to take initiative and handle responsibility, and effective time management skills.
