DESCRIPTION

The Coufal lab studies neuroinflammation and the role of microglia, the inflammatory cell type of the brain, in neurodevelopment, aging, and both idiopathic and genetic neurologic disease using a variety of in vitro and in vivo models and stem cell derived microglia.

Under supervision of the Principal Investigator and in conjunction with postdoctoral researchers, the Staff Research Associate II will participate in a variety of different techniques to isolate nuclei from a variety of different brain types, with a goal of comparing the transcriptomic and epigenetic signature of microglia across sexes and species. These differences may underlie many neurodevelopmental and neurodegenerative diseases and identifying the conserved and divergent aspects of microglia across sex and species will help answer these questions.

Techniques utilized includes nuclei isolation, flow cytometry, and library preparation. Other lab techniques performed in the lab include induced pluripotent stem cell research, differentiation of induced pluripotent stem cells to a variety of cell fates including microglia, neurons, astrocytes, and cerebral organoids, and will learn and utilize general molecular biology experiments including: preparing RNA from cells and tissue, cDNA synthesis and quantitative PCR, performing RNAseq and ChIPseq library preparations, cloning and general molecular biology techniques, western blotting, immunohistochemistry of cells and organoids, and cryostat sectioning. There is the opportunities to learn and master a variety of techniques and to contribute not only technically but also to experimental design as able.

MINIMUM QUALIFICATIONS

  • Theoretical knowledge of molecular biology or cellular biology generally acquired through a Bachelor's degree or an equivalent combination of education and experience.

  • Able to take direction, be detail oriented, follow step-wise protocols, and perform repetitive tasks in a fast-paced laboratory environment. Able to learn new procedures.

  • Ability to work independently and as part of a team, establishing and maintaining good working relationships with P.I., supervisor, postdoctoral scholars, and other members of the research group.

  • Demonstrated ability to communicate effectively in standard English, in writing, electronically, and by spoken word with lab members and vendors.

  • Ability to perform RNA purification, cDNA synthesis, and quantitative PCR assays.

  • Ability to assist lab personnel in tissue culture, including stem cell culture.

  • Knowledge of general molecular biology techniques, and use of common laboratory equipment.

  • Knowledge of laboratory safety including radioactive/biohazard handling & disposal techniques, & the ability to ensure laboratory compliance with required safety regulations.

  • Ability to troubleshoot and perform flow cytometry.

PREFERRED QUALIFICATIONS

  • Experience with flow cytometry.
  • Experience with library preparation.

SPECIAL CONDITIONS

  • Employment is subject to a criminal background check.
  • Must be able to work in environment where biological and chemical hazards are present.
  • Strong preference to candidates with experience in flow cytometry.
  • Strong preference for candidates with experience in library preparation.
  • Must be able to work with human cells including established induced pluripotent stem cells and human embryonic stem cells.
  • Must be willing to variable work schedule to accommodate research needs, including occasional overtime, nights, weekends.
  • Must be technically detailed and careful.
