The electron and light microscopy specialist collaborates with researchers from several laboratories, aids the research group and independently conducts laboratory investigations at NCMIR. Performs and develops both specimen preparation and sectioning techniques in the areas of high resolution transmission and intermediate high voltage electron microscopy, serial block-face scanning microscopy, immunofluorescence, immunocytochemistry, protein labeling, serial sectioning, computer-aided 3-dimensional reconstruction, tomographic reconstruction within thick biological specimen. Develops, modifies and implements freeze fracture techniques, high pressure freezing, 2-photon imaging and confocal microscopy.
Obtains large scale brain mapping data and examines and acquires high quality images of specimens for both data analysis and publication. Assists researchers with the processing and analysis of 3D EM data, including EM volume segmentation and morphometric analysis. Teaches and trains undergraduate students, graduate students, post-docs and collaborators on equipment and laboratory procedures used in confocal and electron microscopy.
Four (4) years of related experience, education/training, OR a Bachelor's Degree in a related area. Related experience includes research in histology, histochemistry, molecular and cellular biology as well as chemistry and biochemistry.
Experience and skill with transmitted light microscopy, laser scanning confocal, transmission electron microscopy, and high voltage electron microscopy.
Experience in the production of high quality light images and electron micorgraphs employing both CCD and Direct Detection devices.
Experience in the production of high quality vibratome sections for confocal microscopy. Familiarity with neuroanatomical landmarks and cellular components in brain slices.
Experience to produce high quality thin and semi-thin sections for electron microscopy with glass and diamond knife.
Experience with high resolution localization labeling techniques (such as colloidal gold etc.) for electron microscopy. Ability to learn new labeling techniques.
Understanding both the theory and practice of various cellular localization labeling techniques for light, confocal, and transmission electron microscopy. Experience with certain cellular localization procedures. Ability to learn new labeling techniques.
Understanding of theory and practice of chemical tissue fixation, high pressure freeze fixation and freeze fracture for localization procedures and ultrastructural procedures used in transmission electron microscopy, confocal microscopy, and light microscopy.
Ability to understand and evaluate scientific literature. This includes all aspects of the research project and to interpret results for scientific publications. Ability to conduct research studies using minimal supervision.
Familiarity and skill with software for processing and creating two-dimensional and three-dimensional images, including use of common visualization tools (e.g., IMOD, AMIRA, etc.)
Skill in providing individual instruction on the use of the transmission electron microscope, confocal microscope, and peripheral support of these instruments.
Excellent written and oral communication skills and ability to juggle multiple projects at once with good organization skills.
- Knowledge of graphing software (e.g., GraphPad prism) and basic understanding of statistical analyses is a plus.
- Basic familiarity with Linux operating system.
